1. Investigating the dissemination of STEM content on social media with computational tools

Abstract

Social media platforms can quickly disseminate STEM content to diverse audiences, but their operation can be mysterious. We used open-source machine learning methods such as clustering, regression, and sentiment analysis to analyze over 1000 videos and metrics thereof from 6 social media STEM creators. Our data provide insights into how audiences generate interest signals(likes, bookmarks, comments, shares), on the correlation of various signals with views, and suggest that content from newer creators is disseminated differently. Abstract

Learning miniaturization science remains challenging due to the non-intuitive behavior of microscale objects and complex assembly approaches. Traditional approaches for creating microsystems require expensive equipment, facilities, and trained staff. To improve as well as democratize microdevice education, we created a new educational activity that enables students to build and test advanced microfluidics by leveraging sticker microfluidics, composed of double-sided dry film adhesive layers. Along with T-mixers and bubble generators, our activity is the first to enable students to build a valve and F-mixer in the classroom setting. This helps emphasize less intuitive aspects of device manufacturing such as the creation of complex 3-dimenstional shapes with layers and layer alignment. In addition, this paper provides the first reported quantitative data on significant improvements in student knowledge and confidence from building and testing several common devices. All 11 students had substantial improvements in conceptual mastery and confidence after the activity. Student responses to a guided reflection highlight how the activity supports a variety of learning needs and preferences. Abstract

This article presents the results of a post-mortem study of various MgO-C and MgO-C-Al pieces that are used in the electric furnaces at the Acerinox factory in Cádiz. From the chemical, thermal, mineralogical and microstructural characterization of the materials prior to use and post-mortem it has been established that, in both types of refractories, the corrosion has lead to the formation of reaction layers with different chemical and mineralogical composition, as a function of temperature and oxygen partial pressure, ranging from the hot to the cold face and strongly depending on the zone of the electric furnace where the piece is situated. Changes in graphite content have been observed in both types of materials by X-ray diffraction. These changes have been quantified by Differential Thermal Analysis. In MgO-C-Al refractories it has been observed the formation of small amounts of C3Al4 inside the pieces and MgAl2O4 in the refractories’ hot face. Keeping in mind the obtained results and the phase equilibrium diagram of Mg-C-Al-O it has been established a corrosion mechanism for both types of materials. It cannot be established by the present results that additions of Al (≈ 2 % in weight) improve the performance of MgO-C materials.Se describen los resultados del estudio post-mortem de diversas piezas de MgO-C y MgO-C-Al utilizadas en los hornos eléctricos de la factoría de Acerinox en Cádiz. De la caracterización química, térmica, mineralógica y microestructural de los materiales sin usar y post-mortem se ha establecido que la corrosión ha dado lugar, en ambos tipos de refractarios, a la formación de capas de reacción con distinta composición química y mineralógica, en función de la temperatura y presión parcial de oxígeno en la pieza, que varían desde la cara caliente a la cara fría y dependen fuertemente de la zona del horno eléctrico en que está situada la pieza. En ambos tipos de materiales se han observado por difracción de rayos X cambios en el contenido de grafito con la distancia a la cara fría. Estos cambios se han cuantificado mediante análisis térmico diferencial. En los refractarios de MgO-C-Al se observa la formación de pequeñas cantidades de C3Al4 en el interior de las piezas y de MgAl2O4 en la cara caliente del refractario. Teniendo en cuenta los resultados obtenidos y los diagramas de equilibrio de fases Mg-C-Al-O se ha establecido el mecanismo de corrosión en ambos tipos de materiales. De los resultados obtenidos no se puede establecer que adiciones de Al (≈ 2% en peso) mejoren el comportamiento en servicio de los materiales de MgO-C.

Development of drug resistance is one of the causes of the therapy failure, being PARP1 (poly ADP-ribose polymerase 1) a relevant protein in the resistance mechanism. In this work, we have functionalized calcium phosphate-based nanoparticles (NPs) with Olaparib (OLA, a PARP-1 inhibitor) in combination with ascorbic acid (AA), a pro-oxidative agent, to enhance their individual effects.Methods: Amorphous Calcium Phosphate (ACP) NPs were synthesized through a biomimetic approach and then functionalized with OLA and AA (NP-ACP-OLA-AA). After evaluation of the loading capacity and release kinetic, cytotoxicity, cell migration, immunofluorescence, and gene expression assays were performed using pancreatic tumor cell lines. In vivo studies were carried out on tumors derived from the PANC-1 line in NOD SCID gamma (NSG) mice.Results: NP-ACP-OLA-AA was loaded with 13%wt of OLA (75% loading efficiency) and 1% of AA, respectively. The resulting dual nanosystem exhibited a gradual release of OLA and AA, being the latter protected from degradation in solution. This ensured the simultaneous availability of OLA and AA for a longer period, at least, during the entire time of in vitro cell experiments (72 hours). In vitro studies indicated that NP-ACP-OLA-AA showed the best cytotoxic effect outperforming that of the free OLA and a higher genotoxicity and apoptosis-mediated cytotoxic effect in human pancreatic ductal adenocarcinoma cell line. Interestingly, the in vivo assays using immunosuppressed mice with PANC-1-induced tumors revealed that NP-ACP-OLA-AA produced a higher tumor volume reduction (59.1%) compared to free OLA (28.3%) and increased the mice survival.Conclusion: Calcium phosphate NPs, a highly biocompatible and biodegradable system, were an ideal vector for the OLA and AA co-treatment in PC, inducing significant therapeutic benefits relative to free OLA, including cytotoxicity, induction of apoptosis, inhibition of cell migration, tumor growth, and survival.Graphical Abstract: Keywords: nanomedicine, calcium phosphate nanocarriers, Olaparib, ascorbic acid, synergistic effect, pancreatic cancer

14. In vitro testing of host-targeting small molecule antiviral matriptase/TMPRSS2 inhibitors in 2D and 3D cell-based assays

Abstract

The outbreak of coronavirus disease 2019 (COVID-19) pandemic strongly stimulated the development of small molecule antivirals selectively targeting type II transmembrane serine proteases (TTSP), required for the host-cell entry of numerous viruses. A set of 3-amidinophenylalanine derivatives (MI-21, MI-472, MI-477, MI-485, MI-1903 and MI-1904), which inhibit the cleavage of certain viral glycoproteins was characterized in 2D and 3D primary human hepatocyte models on collagen- and Matrigel-coating using a CCK-8 assay to evaluate their cytotoxicity, a resorufin-based method to detect redox imbalances, fluorescence and ultrafiltration experiments to evaluate their interactions with human serum albumin (HSA) and α-acidic glycoprotein (AGP), and luminescence measurement to assess CYP3A4 modulation. For elucidation of selectivity of the applied compounds towards matriptase, transmembrane serine protease 2 (TMPRRS2), thrombin and factor Xa (FXa) Ki values were determined. It was proven that cell viability was only deteriorated by inhibitor MI-1903, and redox status was not influenced by administration of the selected inhibitors at 50 µM for 24 h. MI-472 and MI-477 formed relatively stable complexes with AGP. CYP3A4 inhibition was found to be strong in PHHs exposed to all inhibitors with the exception of MI-21, which seems to be a promising drug candidate also due to its better selectivity towards matriptase and TMPRSS2 over the blood clotting proteases thrombin and FXa. Our in vitro pharmacokinetic screening with these inhibitors helps to select the compounds with the best selectivity and safety profile suitable for a further preclinical characterization without animal sacrifice.

17. Explaining the Relation between Early Mind-Mindedness and Children's Mentalizing Abilities: The Development of an Observational Preschool Assessment

Abstract

The aim of this research was to develop a new observation-based measure for assessing caregivers' mind-mindedness in the preschool years and investigate whether this measure could explain the link between mothers' early appropriate mind-related comments and children's later mentalizing abilities. The new measure was developed using a sample of mothers and 44-month-olds (N = 171), characterizing mind-mindedness in terms of (a) solicited child involvement, (b) adaptive communication, and (c) internal state talk. These indices were positively related to established assessments of mind-mindedness at 8, 44, and 61 months. Positive associations were also observed with children's later mentalizing abilities. The new measure of mind-mindedness did not, however, mediate the relation between mind-mindedness in the first year of life and children's mentalizing abilities.

23. Telehealth for geriatric post-emergency department visits to promote age-friendly care

Abstract

Objective: To describe a feasibility pilot study for older adults that addresses the digital divide, unmet health care needs, and the 4Ms of Age-Friendly Health Systems via the emergency department (ED) follow-up home visits supported by telehealth. Data Sources and Study Setting: Data sources were a pre-implementation site survey and pilot phase individual-level patient data from six US Department of Veterans Affairs (VA) EDs. Study Design: A pre-implementation survey assessed existing geriatric ED processes. In the pilot called SCOUTS (Supporting Community Outpatient, Urgent care & Telehealth Services), sites identified high-risk patients during an ED visit. After ED discharge, Intermediate Care Technicians (ICTs, former military medics), performed follow-up telephone, or home visits. During the follow-up visit, ICTs identified "what matters," performed geriatric screens aligned with Age-Friendly Health Systems, observed home safety risks, assisted with video telehealth check-ins with ED providers, and provided care coordination. SCOUTS visit data were recorded in the patient's electronic medical record using a standardized template. Data Collection/Extraction Methods: Sites were surveyed via electronic form. Administrative pilot data extracted from VA Corporate Data Warehouse, May-October 2021. Principle Findings: Site surveys showed none of the EDs had a formalized way of identifying the 4 M "what matters." During the pilot, ICT performed 56 telephone and 247 home visits. All home visits included a telehealth visit with an ED provider (n = 244) or geriatrician (n = 3). ICTs identified 44 modifiable home fall risks and 99 unmet care needs, recommended 80 pieces of medical equipment, placed 36 specialty care consults, and connected 180 patients to a Patient Aligned Care Team member for follow-up. Conclusions: A post-ED follow-up program in which former military medics perform geriatric screens and care coordination is feasible. Combining telehealth and home visits allows providers to address what matters and unmet care needs. Dark matter could be made up of dark photons, massive but very light particles whose interactions with matter resemble those of usual photons but suppressed by a small mixing parameter. We analyze the main approaches to dark photon interactions and how they can be applied to direct detection experiments which test different ranges of masses and mixings. A new experiment based on counting dark photons from induced atomic transitions in a target material is proposed. Abstract

State-of-the-art technology for pulse counter electronics offers an important range of commercial devices, but such systems are usually expensive due to the complex logic used for this task. The use of counting electronics in conjunction with photon counters can be used, for example, to perform experimental tests in Quantum Optics and Quantum Information Science. Here, we present the development and implementation of a low cost module for multiphoton coincidence statistics with detection windows of a few nanoseconds. The module consists of an array of logic gates, with a frequency operation of 250 MHz that corrects and amplifies the detectors signal. The device characterisation was done by means of detection of Transistor-Transistor Logic (TTL) signals retrieved from a signal generator, and implemented in an optical setup. The detected output signals (TTL pulses) were analysed and stored in a computer by means of a Field Programmable Gate Array (FPGA). Abstract

Educating new students in miniaturization science remains challenging due to the non-intuitive behavior of microscale objects and specialized layer-by-layer assembly approaches. In our analysis of the existing literature, we noted that it remains difficult to have low cost activities that elicit deep learning. Furthermore, few activities have stated learning goals and measurements of effectiveness. To that end, we created a new educational activity that enables students to build and test microfluidic mixers, valves, and bubble generators in the classroom setting with inexpensive, widely-available materials. Although undergraduate and graduate engineering students are able to successfully construct the devices, our activity is unique in that the focus is not on successfully building and operating each device. Instead, it is to gain understanding about miniaturization science, device design, and construction so as to be able to do so independently. Our data show that the activity is appropriate for developing the conceptual understanding of graduate and advanced undergraduate students (n = 57), as well as makes a lasting impression on the students. We also report on observations related to student patterns of misunderstanding and how miniaturization science provides a unique opportunity for educational researchers to elicit and study misconceptions. More broadly, since this activity teaches participants a viable approach to creating microsystems and can be implemented in nearly any global setting, our work democratizes the education of miniaturization science.
